Jika sebelumnya Ducati Panigale dibekali mesin berkapasitas 899 cc, untuk generasi terbaru ini ditingkatkan menjadi 955 cc. Meski kapasitas mesin naik, Ducati mengklaim kalau tingkat emisi yang dihasilkan lebih rendah dari Panigale sebelumnya dan lolos standar EURO 4.
Ducati Panigale 959 bisa dibilang gabungan dari Panigale 899 dan 1299. Pada sektor roda mengaplikasi pelek cast wheel palang 10, lalu rem menggunakan Brembo Monoblock dan swing arm aluminium dari panigale 899. Untuk suspensinya menggunakan Showa di depan dan Sachs di belakang.
